Depends on the situation. Well lit and shooting on the fly, changing focal lengths as I go, I'm relying on the auto focus.

Camera on a tripod - moody light - everything locked... I know exactly when I'm pushing the shutter and everything is pre-set - and there's even the slightest hint that there may not be enough contrast out there for the camera to lock in on - I'll manually focus ahead of time and leave it.

My opinion is that you need to go with the situation - that's why you have options. Test the camera before the train comes. Is it hunting? If so, then preset the focus manually.
